There is no single required volume — the diluent sets the concentration, not the amount of Semax in the vial. A 5mg vial (5 mg) gives 5 mg/mL with 1 mL of bacteriostatic water, 2.5 mg/mL with 2 mL, and 1.67 mg/mL with 3 mL. More diluent makes each aliquot a larger, more precisely measurable volume.
Concentration is the lyophilized mass divided by the diluent volume. A 30mg Semax vial reconstituted with 2 mL of bacteriostatic water is 15 mg/mL (15,000 mcg/mL). Semax is supplied in 5mg, 10mg, 30mg vials; the chart on this page lists each size at 1, 2 and 3 mL.
A U-100 graduated syringe has 100 marks per milliliter, so each mark is 0.01 mL. At 2.5 mg/mL (a 5mg vial with 2 mL) each mark holds 25 mcg; multiply the mcg-per-mark figure by the number of marks to find the mass in a measured volume.
